How does the combination of sodium/potassium sucrose octasulfate and NOCMC maintain skin moisture balance?

Sucrose octasulfates are a group of compounds with the structure C12H14O35S8-X8 (left figure), where X can represent elements such as K, Na, Al, etc. Sucrose octasulfate aluminum salt was first used in the treatment of ulcers and plays an important role in the treatment and prevention of chemotherapy-induced stomatitis. In 2015, with the listing of Irinotecan liposomes (right) of Taiwan, China Zhiqing Company in the United States, another new variety was added to the liposome industry. Sucrose sulfate, as a new excipient, came into the eyes of pharmaceutical colleagues. Irinotecan liposome was approved by FDA to treat metastatic pancreatic cancer in 2016, which is a typical application case of sucrose octasulfate.

Recent research has invented a topical composition using sodium or potassium sucrose octasulfate and N, O-carboxymethyl chitosan (NOCMC) as active ingredients. This composition can be used to maintain and restore skin water balance and can be applied in dermatology and aesthetics.

The balance of moisture is crucial for maintaining the integrity, health, and good condition of the skin. Transepidermal dehydration (TEWL) is a physiological phenomenon defined as the transport of water through the skin through passive diffusion, including continuous diffusion of water from the deepest to the outermost layers of the skin. External moisturizers, such as a sealing layer formed by mineral fat covering the skin, can reduce the movement of water, slow down the TEWL of the skin, increase hydration in the skin, and maintain the skin's moisture balance. However, moisturizers can sometimes cause changes in the homeostasis of skin surface cells and microorganisms, increasing the risk of bacterial and fungal infections.

It has been proven that the combination of Ν, Ο - carboxymethyl chitosan and sodium or potassium sucrose octasulfate has a hydrophilic synergistic effect. Topical combinations with both as active ingredients can effectively absorb and retain moisture, slow down the loss of water through the epidermis, and maintain and restore the skin's water balance. The composition of the present invention is particularly suitable for the fields of pediatrics and geriatric diseases.

The sodium or potassium salt of sucrose octasulfate in this composition exhibits high water solubility and no local or systemic toxicity. Sucrose octasulfate sodium/potassium salt is easily available in the market. At present, Aiweituo Company's sodium sucrose octasulfate and potassium sucrose octasulfate have been launched in China and are planned to be registered with CDE. In the composition, N, O-carboxymethyl chitosan (NOCMC) is an important water-soluble chitosan derivative, which has many medical effects, such as promoting wound healing, hemostasis, inhibiting scar, analgesia and bacteriostasis. It has good biocompatibility and biodegradability, and is widely used in hydrogels and wound healing biomaterials.

In this composite component, N, O-carboxymethyl chitosan is present in the composition in weight percentages ranging from 0.010% to 3%. The percentage of sodium or potassium sucrose octasulfate in the composition ranges from 0.100% to 1.0%.

In addition, the composition of the invention can contain other active ingredients commonly used in dermatology and cosmetology, and can be prepared into lotion, gel, lotion, cream, etc. by conventional methods.
