How to ensure the reproducibility of ELISA experiments?

1、 Standard product handling and sample addition specifications

Standard processing: Freeze dried standard samples need to be dissolved in a matching diluent, vortex mixed, and then diluted in a gradient (such as 640pg/ml to 0pg/ml) to avoid repeated freezing and thawing.

Sample specification:

Use a calibrated pipette and slowly inject the sample with the pipette tip attached to the wall at a 45 ° angle to avoid bubbles and liquid sticking to the wall.

Control the sampling speed (to be completed within 5 minutes), and ensure that the inter hole error is less than 5%.

2、 Incubation control and washing optimization

Incubation control: Incubate at a constant temperature of 37 ℃, seal the plate with a membrane to prevent evaporation, and use a shaking instrument to assist in mixing.

Washing optimization:

Wash solution prepared and used immediately (diluted 1:20), fill each well and let it stand for 30 seconds, repeat 5 times.

Avoid residue when shooting dry, and focus on checking the edge holes.

3、 Sealing reinforcement and reagent balance

Sealing enhancement: Seal with 1% BSA or gelatin for 1.5 hours to reduce non-specific binding.

Reagent equilibrium:

The reagent kit should be equilibrated at room temperature for 15-30 minutes, and the substrate should be stored away from light.

After opening, the enzyme-linked immunosorbent assay (ELISA) plate should be stored in a sealed bag to prevent moisture.

4、 Sample processing and pipette calibration

Sample processing:

Centrifuge the serum sample and package it separately to avoid repeated freezing and thawing; High concentration samples require pre dilution.

Hemolytic or lipid blood samples require ultrafiltration treatment.

Pipette calibration: Calibrate the pipette monthly and check the sealing before adding samples.

5、 Incubation records and batch management

Incubation record: Use a timer to control the incubation time and avoid human error.
