Process flow of eye drops filling production line
Date: 2025-12-24Read: 9
Eye drops, as sterile preparations that directly act on the eyes, have extremely strict requirements for cleanliness, sterility assurance, and filling accuracy in their production process. The following are the core process flow and key technical points of the eye drop filling production line:
1、 Core process flow
1. Raw material preparation and container handling
The raw materials must meet pharmaceutical standards, and the active ingredients must undergo stability testing.
Containers (such as plastic eye drops) need to be cleaned and sterilized: soaked in 0.5% sodium nitrate solution, rinsed with pure water, and sterilized by dry heat.
The three section bottle (integrated design of bottle body, inner stopper, and bottle cap) can be used directly, but microbiological indicators need to be sampled and tested.
2. Liquid preparation and filtration
Choose the preparation method based on drug stability: Thermally stable drugs can be prepared centrally, while thermosensitive drugs need to be prepared and sterilized on site.
The filtration uses a 0.22 μ m microporous membrane for sterilization, ensuring the sterility of the drug solution.
The osmotic pressure should be controlled between 0.6% -1.5% NaCl equivalent, and the pH value should be between 5.0-9.0.
3. Aseptic filling
Filling environment: It needs to be operated in a Class 100 clean area (under a Class A laminar flow hood), with a background environment of a Class 10000 clean area (Class C).
Filling method:
Traditional filling: The medicine is injected into the container through a separation tank, positioned by a camera, and the bottle cap is tightened by rotating the spindle to ensure sealing.
Blow sealing (BFS) technology: Plastic raw materials are blow molded into bottles, filled with medication, and sealed in an integrated manner in a sterile environment, without the need for additional sterilization.
Accuracy control: The filling volume error should be controlled within ± 1%, and the sealing of the bottle cap should be verified through torque testing.
4. Sterilization and Quality Inspection
Sterilization method: Heat stable drugs are sterilized by hot pressing, while thermosensitive drugs are sterilized by aseptic operation or filtration.
Quality inspection:
Clarity detection: Check for visible foreign objects in the medicine solution using a lamp inspection machine.
Aseptic testing: membrane filtration or direct inoculation methods are used to ensure no microbial contamination.
Osmotic pressure and pH value: detected by instruments, in compliance with pharmacopoeia standards.
Sealing test: Negative pressure method or staining method is used to test the sealing of containers.
5. Packaging and Storage
Labeling and packaging must be completed in a clean environment, and label information must comply with regulatory requirements.
Stability testing is required before the finished product is stored to ensure stable quality within its validity period.
2、 Key technical points
1. Cleanliness control
The filling area needs to achieve Class A cleanliness (ISO 5) and a background environment of Class B (ISO 7).
Adopting MEGAcel ® Efficient filter, nano scale ePTFE membrane can reduce operating resistance by 50% and eliminate the risk of fiber shedding.
2. Aseptic assurance technology
BFS technology: By integrating blow molding, filling, and sealing, intermediate pollution is reduced without the need for preservatives.
CIP/SIP system: online cleaning and sterilization to ensure equipment sterility.
3. Automation and Intelligence
The filling line is equipped with a SCADA system to monitor real-time parameters such as temperature, pressure, pH value, etc.
Camera positioning and torque control system ensures filling accuracy and sealing.
4. Environmental Protection and Safety
Some filters use recyclable materials, which meet the requirements of low-carbon production.
Hormonal and non hormonal products need to be produced separately to avoid cross contamination.
3、 Industry Frontiers and Challenges
1. The popularization of BFS technology
Domestic enterprises have gradually adopted BFS production lines to produce single dose eye drops without preservatives.
Challenge: The problem of electrostatic adsorption of dust particles on plastic bottles needs to be solved through raw material pretreatment or environmental control.
2. Risk assessment of co production
Feasibility assessment and risk control measures need to be developed for the co production of hormone and non hormone products.
It is recommended to use specialized equipment or produce in different time periods to reduce pollution risks.
3. Improvement of cleanliness of packaging materials
The cleanliness level of ophthalmic drug packaging materials should match the production of the formulation to avoid introducing contamination.
The airtightness of the joint between the bottle body and the inner stopper of the three section bottle needs to be further optimized.

The process flow of the eye drop filling production line integrates aseptic technology, clean engineering, and automation control. The core lies in ensuring the safety and effectiveness of each eye drop through strict environmental control, advanced filling technology, and comprehensive quality testing. With the popularization of BFS technology and intelligent monitoring systems, the production of eye drops will develop towards higher efficiency and lower pollution in the future.
