The separation column is the core of chromatographic analysis, and the protective column can protect the separation column and significantly improve its service life. When it is necessary to replace the protective column and chromatographic column with new ones, many macaroni do not know how to operate them. Here is an introduction:
1. Replace the protective pillar with a new one
2. Replace the chromatography column with a new one

Kind Reminder

Course content fragment:

1、 Replace the protective pillar with a new one
It is recommended to always use protective columns during work. The selection of protective columns can be consulted with Wantong agents or after-sales personnel.
The new protective column has been filled with solution and sealed at both ends with plugs or caps. Before use, it is necessary to confirm that the solution is miscible with the rinsing solution used.
After the instrument is put into operation, the replacement of the protective column can be carried out according to the following steps:
1. Connect the protective column
Connect the protective column according to the steps shown in the diagram below:
(1) Disconnect the two-way or old protective column from the chromatographic column
Disconnect the input capillary tube from the old protective column or two-way connector;
(2) Prepare a new protective pillar
Remove the plugs and sealing caps from both ends of the new protective column;
(3) Connect the new protective pillar
Connect the protective column to the short PEEK pressure screw at the input capillary end;
Note:When connecting the protective column, it is necessary to use it correctly according to the specified flow direction.

2. Rinse the protective column
(1) Place the large mouthed cup under the outlet of the protective column;
(2) Start manual control in MagIC Net and select high-pressure pump: Manual - Manual Control - Pump
- Set flow rate: in accordance with the chromatographic column instructions
- Click to start

(3) Rinse the protective column with rinsing solution for about 5 minutes;
(4) Stop the high-pressure pump again in manual control of MagIC Net: off.

2、 Replace the chromatographic column with a new one
As the core of chromatographic analysis, the chromatographic column separates different components and is equipped with a chip that stores its process specifications and historical records (such as operation, working hours, injection times, etc.).
The new separation column is filled with solution and both ends are sealed with plugs. Before using the column, please confirm that the solution is miscible with the solution being used.
For instruments that have already been put into operation, a new separation column can be connected according to the following steps and corresponding diagram.
1、 取出塞子
Remove the plugs from both ends of the separation column:

2. Install separation column inlet
Twist the protective column directly onto the inlet end of the separation column
If the protective column is connected to the separation column through a capillary tube, PEEK (polyetheretherketone) pressure screws (6.2744.070) need to be used to tighten the capillary tube to the inlet end of the separation column.

3. Rinse the separation column
1) Place the large mouthed cup under the outlet of the separation column;
2) Start manual control in MagIC Net and select high-pressure pump: Manual - Manual Control - Pump
- Set flow rate: in accordance with the chromatographic column instructions
- Click to start

3) Rinse the separation column with rinsing solution for about 10 minutes;
4) Restarting the high-pressure pump in manual control of MagIC Net: Off

4Remove two or old chromatography columns and protective columns
Remove the two-way or old protective column and chromatographic column connected to the output capillary end.

5Install the outlet of the separation column
Secure the column output capillary tube to the upper end of the separation column using a short PEEK (polyetheretherketone) pressure screw (6.2744.070).

6. Install separation column
Insert the separation column with chip into the column clamp until you hear the sound of the card.

At this point, MagIC Net recognizes the separation column. After the installation of the separation column is completed, the system balance can be entered.
