Detailed explanation of the causes and solutions of UV visible spectrophotometer malfunctions
Date: 2025-12-01Read: 8
As a fundamental equipment in the field of physical and chemical analysis, UV visible spectrophotometers are widely used in various fields such as pharmaceutical testing, environmental monitoring, food analysis, and material research due to their high sensitivity and wide measurement range advantages. The device has a precise structure and involves multiple systems such as optics, circuits, and mechanics. Any abnormality in any link may affect the reliability of the detection results. This article combines laboratory practical experience to systematically sort out the common types of faults, deep causes, and targeted solutions of UV visible spectrophotometers, providing efficient troubleshooting ideas for technical personnel.
Startup failure is the primary obstacle to device operation, mainly manifested as no response from the instrument after power on, indicator lights not turning on, or automatic shutdown after startup. From the perspective of the cause of the malfunction, the first step is to investigate the basic power supply system: poor contact of the power cord, loose sockets, or fluctuations in power supply voltage (deviating from the rated range of 220V ± 5%) are the causes, which can be solved by replacing the socket, re plugging the power cord, or using a voltage regulator. If the power supply is normal but still cannot be started, the instrument fuse (usually located at the power interface) needs to be checked. If it is blown, replace it with a fuse of the same specification; If the fuse is intact, it may be a damaged power module and the manufacturer needs to be contacted for professional repair. Some instruments automatically shut down after startup, which is often caused by poor heat dissipation. Cleaning the debris in the instrument ventilation port and ensuring the normal operation of the cooling fan can restore it.
Baseline anomalies are the core issues that affect detection accuracy, manifested as baseline drift, excessive noise, or irregular fluctuations. The main reasons for baseline drift include insufficient preheating time of the instrument (usually requiring 30-60 minutes of preheating, especially in the ultraviolet band), excessive temperature fluctuations in the laboratory (exceeding the constant temperature range of 20-25 ℃), and unclean or scratched colorimetric dishes. The solution is to extend the preheating time, turn on the laboratory air conditioning constant temperature, clean the colorimetric dish with alcohol or deionized water and wipe it clean, and replace it with a new one if necessary. Excessive noise is related to aging of the light source, contamination of the monochromator, or decreased sensitivity of the detector. If there is obvious noise in the ultraviolet region, it may be due to the expiration of the deuterium lamp's lifespan (usually 1000-2000 hours), and the deuterium lamp needs to be replaced; Monochromator pollution can be cleaned by the manufacturer's engineer by disassembling and cleaning the optical components; The detector malfunction needs to be professionally detected and replaced. In addition, poor grounding of the instrument can also cause baseline fluctuations, ensuring that the instrument grounding circuit is firmly connected is sufficient.
Abnormal measurement data is the most intuitive manifestation of a malfunction, including inaccurate absorbance values, poor repeatability, or excessive deviation of measurement results from standard values. Common reasons for inaccurate absorbance values include inaccurate wavelength, poor matching of colorimetric dishes, or sample preparation issues. When there is a wavelength shift, a standard filter (such as holmium glass) needs to be used for wavelength calibration, which can be adjusted through the instrument's built-in calibration function; Differences in thickness or transmittance of colorimetric dishes can lead to errors. Therefore, colorimetric dishes of the same batch and specification should be selected and paired for testing before use. Poor repeatability is often caused by improper operation, such as inconsistent injection volume, incorrect placement of colorimetric dishes, and uneven sample shaking. Standardizing the operation process can improve this issue; If there is still no improvement, it is necessary to check whether the injection system is blocked or the peristaltic pump flow rate is unstable, clean the injection pipeline or calibrate the peristaltic pump. If the measurement result deviation is too large, it is necessary to check whether the sample has deteriorated, whether the standard solution has expired, and prepare the sample and standard solution again before testing.
The malfunction of the light source system directly leads to the malfunction of the instrument, and different light source malfunctions manifest differently. The common fault of deuterium lamp (ultraviolet region) is that it cannot light up or the luminous intensity is insufficient: when it cannot light up, first check whether the wiring of the deuterium lamp is loose, and if it is loose, plug and unplug it again; If the wiring is normal, it may be a fault in the deuterium lamp trigger circuit and requires professional repair. Insufficient luminous intensity is often caused by the aging of deuterium lamps. The self checking function of the instrument can be used to check the usage time of deuterium lamps, and they can be replaced when their lifespan is reached. The malfunction of tungsten lamp (visible area) is similar. If the brightness is dim or flashes after lighting up, in addition to replacing the bulb, it is necessary to check whether the lamp holder is in good contact, clean the oxide of the lamp holder, and reinstall it. Some instrument light source switching faults are mostly caused by switching motor faults or control circuit problems. Contact the manufacturer for repair.
The malfunction of the operating interface affects the normal operation of the device, manifested as unresponsive touch screens, malfunctioning buttons, or displaying garbled characters. When the touch screen does not respond, restart the instrument first to eliminate software lag. If it is ineffective, calibrate the touch screen (through the instrument calibration menu operation); Button failure is often caused by aging or poor contact. Clean the button contacts or replace the button panel. Displaying garbled characters is usually a system program malfunction, which can be resolved by the instrument's built-in factory reset function. If it cannot be restored, the system program needs to be reinstalled. In addition, if data cannot be transmitted after connecting to the computer, it is necessary to check whether the USB or serial port connection cable is damaged, reinstall the driver program or replace the data cable.
Troubleshooting needs to follow“The principle of "from simple to complex": When repairing components such as circuits and light sources, the power must be cut off first to avoid electric shock or damage to the components; Optical components (such as monochromators and reflectors) are delicate and fragile, and should not be disassembled or cleaned by oneself. They must be operated by professional personnel. Daily maintenance is the key to reducing failures: establishing equipment usage records, recording light source usage time, calibration status, and fault handling records; Clean the colorimetric dish, wipe the surface of the instrument and ventilation openings weekly; Monthly inspection of light source brightness and baseline checks; Calibrate wavelength and absorbance using standard filters every quarter; When not in use for a long time, turn on the power regularly (once a month, 30 minutes each time) to prevent components from getting damp and aging.
The troubleshooting of UV visible spectrophotometers requires a combination of optical principles and practical experience to accurately locate the cause through step-by-step investigation. In daily use, standardizing operating procedures and performing regular maintenance can significantly reduce the incidence of malfunctions. Technicians need to be familiar with the structure of the instrument and the working principles of each system, accumulate experience in fault handling, ensure that the equipment is always in a stable operating state, and provide reliable guarantees for accurate analysis.
