1. Only equipped with 10mL of composite neutralizing eluent; Required but not provided:

1)Sterile specification plate, sterile material sampling swab, swab head wetting solution;

2)Tryptone soy agar, nutrient agar, sterile culture dish - used for pouring method;

3)Improve pouring culture bottles, etc.

Used for environmental monitoring: microbial limit testing of object surfaces and hand hygiene. Wash the bacteria off the sampling swab and neutralize any residual disinfectant. No bacterial growth function, not part of the culture medium.

【 Principle of Inspection 】

Quantitative sampling uses various neutralizing agents contained in the product to neutralize residual disinfectants on the sample, in order to release the inhibition of residual disinfectants on microorganisms, facilitate the cultivation and revival of residual microorganisms, and avoid missed detections.

The composite neutralizing agent contained in the liquid can neutralize various disinfectants, including iodine, chlorine containing disinfectants, peroxides, quaternary ammonium (benzalkonium bromide), aldehydes, biguanides (chlorhexidine), phenols, and alcohols. Therefore, there is no need to consider matching with the type of disinfectant when using this product, and there is no difficulty in choosing.

[Appearance]

Opaque, milky white to amber in color, with slight precipitation after prolonged storage (does not affect filtration, should be shaken and mixed before use).

Storage conditions and expiration date

Avoid light, at room temperature (10-30 ℃), with a shelf life of one year. Please refer to the label for details. Open the lid and use it immediately, open the bag and use it immediately.

1. Specimen type:

lSurface sampling refers to swab heads used to collect samples from surfaces that have been disinfected with disinfectants within a specified area;

lSampling of curved surfaces with both hands, using swab heads for disinfected medical staff to sample curved surfaces with both hands;

2. Time requirement: Collect specimens within 4 hours after disinfection and deliver them to the laboratory for experimentation within 2 hours. If experiments cannot be conducted in a timely manner, they should be

Refrigerate at 2-8 ℃ and process within 24 hours.

1. Microbial limit test on the surface of objects (non contact plate method), surface after disinfection with disinfectant

1.1 sampling

1.1.1Sampling timing: Sampling within 4 hours after disinfection

1.1.2Sampling area: When the sampling area is less than 100 cm2, collect all surfaces; When the sampling area is ≥ 100cm2, use a specification plate to collect 100cm2

Surface area of the object.

1.1.3Collection method:

1)Place a 5cm × 5cm sterilized specification plate on the surface of the object to be tested, soak the sampling swab head with sterile phosphate buffer solution prepared in the bag, apply it horizontally and vertically back and forth 5 times inside the specification plate, and then rotate the swab accordingly, continuously sampling 4 specification plates with an area of 100cm2;

2)Before use, gently shake and mix well to neutralize the eluent;

3) Insert the swab into the test tube and place the break point on the swab rod near the edge of the test tube mouth. Grasp the test tube with your left hand and the swab rod with your right hand, bend it about 45-60 degrees and break it, so that the swab head falls into the test tube. Discard the swab rod held in your right hand, tighten the test tube cap containing the swab head in a timely manner, and send it for testing in a timely manner; The operation diagram is as follows:

4)Small objects such as door handles with irregular shapes or less than 100 cm2 should be wiped with a moist swab head to collect the entire surface, without the need for a specification plate.

1.2 Elution: Shake the test tube containing the sampling swab head thoroughly on a vortex oscillator to elute the sample for at least 30 seconds, then immediately remove the swab head and discard it to ensure the reproducibility and accuracy of the results;

1.3 Vaccination:

1) Operating environment: At least 100000 level clean environment to prevent pollution, such as biosafety cabinets;

2)Take 1mL from 10mL of the eluted sample and the eluent, and perform colony counting using the pouring method (see Appendix A);

1.4 Cultivation: Cultivation: 35 ℃± 2 ℃, atmospheric environment, 48 hours;

1.5 Counting and Calculation

lWhen sampling 100 cm2, CFU/cm2=colony count x 10 * ÷ 100cm ²;

lWhen sampling<100 cm2, CFU/piece=colony count x 10 *;

Note: * Take 1mL of the neutralizing eluent from 10mL of the eluted sample and pour it for counting, i.e. dilute 10 times.

1.6 Standards and Interpretation

1.6.1The microbial limit results on the surface of the object after disinfection with disinfectant should meet the requirements of Table 1

Table 1. Surface hygiene standards for objects

Environmental category

Average colony count on the surface of an object (CFU/cm2)

Class I environment

≤ 5.0

Class II environment

≤ 5.0

Class III environment

≤ 10.0

Class IV environment

≤ 10.0

lClass I environment refers to medical and treatment facilities that use clean air technology, divided into clean operating rooms and other clean areas;

lClass II environment, including non clean operating rooms, delivery rooms, catheterization rooms, hematology wards, burn wards and other protective isolation wards, intensive care units, neonatal rooms, etc;

lClass III environment, including mother and baby rooms, inspection and packaging sterilization areas, sterile item storage areas in disinfection supply centers, hemodialysis centers (rooms), and other general inpatient wards;

lClass IV environment, which is the general outpatient (emergency) clinic and its examination and treatment room, infectious diseases clinic and ward.

1.6.2Result explanation: If the calculated result meets the standard, it is qualified; On the contrary, it is unqualified.

2. Medical staff's hand hygiene check, hands disinfected with disinfectant (wearing gloves)

2.1 sampling

2.1.1Sampling timing: After practicing hand hygiene, sample before coming into contact with the patient or engaging in medical activities;

2.1.2Sampling method: Soak the sampling swab head in sterile phosphate buffer solution prepared in the bag and rub it back and forth on the curved surface of both fingers from finger to finger end twice (with one hand rubbing an area of about 30 cm2), then rotate the sampling swab head accordingly, insert the swab head into the test tube, and the break point of the swab is against the edge of the test tube mouth. Hold the test tube with your left hand and the swab rod with your right hand, bend it about 45-60 degrees, and break it. The swab head falls into the test tube, discard the swab rod held by your right hand, tighten the test tube cap containing the swab head in a timely manner, and send it for testing in a timely manner;

2.2 Elution: Shake the test tube containing the sampling swab head thoroughly on a vortex oscillator to elute the sample for at least 30 seconds, then immediately remove the swab head and discard it to ensure the reproducibility and accuracy of the results;
