Pierce IP lysis buffer can effectively lyse cultured mammalian cells from well plate cells and cells granulated in suspension culture medium. This lysis buffer has been optimized for various pull-out assays and precipitation detection, and is compatible with many other applications, including Thermo Scientific Pierce BCA and 660 nm protein assays, protein purification, and various detection methods such as ELISA and Western blotting.
Pierce IP lysis buffer consists of 25 mM Tris HCl pH 7.4, 150 mM NaCl, 1 mM EDTA, 1% NP-40, and 5% glycerol. This buffer does not contain protease or phosphatase inhibitors; But if necessary, various inhibitors can be added before use, such as Thermo Scientific Halt protease inhibitor mixture or phosphatase inhibitor mixture, to prevent protein hydrolysis and maintain protein phosphorylation.
