1、 Preparation before extraction with a separating funnel
Check whether the lid and plug of the separating funnel are tightly selected, and select a separating funnel with a total volume of the extracted solution and extractant that is more than twice as large. The method of checking whether the separating funnel leaks is usually to add a certain amount of water, shake it, and see if there is a leak. Warning: A: The lid cannot be coated with oil. B: Do not use the separating funnel with a leak to ensure operation**
II Add extraction solution
Pour the extracted solution and extractant separately into the upper opening of the separating funnel, cover the lid, and select the extractant based on the solubility of the extracted substance in the solvent. It should also be easy to separate from the solute and preferably use low boiling point solvents. Substances with high water solubility can be extracted with benzene or ether, while substances with low water solubility can be extracted with petroleum ether; The highly water-soluble liquid is divided into two phases using ethyl acetate. III Extraction oscillation
1. Oscillate the separating funnel to ensure full contact between the two-phase liquid layers. The oscillation operation generally involves tilting the separating funnel so that the upper opening of the funnel is slightly downward, and mixing the liquid into an emulsion. Note that: A. Apply great force during oscillation and prevent liquid leakage. 2. After the extraction oscillation is completed, release the gas and oscillate. Keep the separating funnel tilted, unscrew the valve, release steam or gas, and balance the internal and external pressures. Attention to gas release: A. Remember to tilt the upper opening of the liquid funnel downwards when releasing gas, and there should be no liquid at the lower opening. 3. Let the separating funnel stand still in the fixture to separate the unstable emulsion. In general, the liquid should be left to stand for about 10 minutes. For those that are difficult to stratify, it should be left to stand for a longer period of time. The liquid should be divided into two clear layers. Note: during extraction. Especially when the solution is alkaline, emulsification often occurs, which affects separation. The methods to destroy emulsification include:
a. Gently swirl the funnel to accelerate layering .
If emulsification occurs due to partial solubility of two solvents (water and organic solvents), a small amount of electrolyte (such as sodium chloride) can be added to disrupt it through salt precipitation. If emulsification occurs due to a small density difference between the two phases, electrolyte can also be added to increase the density of the aqueous phase
d. If emulsification occurs due to the alkaline nature of the solution, a small amount of dilute hydrochloric acid can be added or filtered to eliminate it
According to different situations, ethanol, sulfonated castor oil, etc. can also be added to eliminate emulsification
Maintenance and upkeep:
1. Proper use of instruments and keeping them in good working condition can prolong their lifespan.
2. During continuous operation, the instrument should be regularly inspected every three months to check for any water droplets, dirt, or other debris falling onto the motor and control components; Check fuses, control components, and fastening screws.
3. The bearings of the transmission part have been filled with an appropriate amount of lubricating grease No. 1 calcium sodium base before leaving the factory. During continuous operation of the instrument, lubricating grease should be added every six months, with a filling amount of about 1/3 of the bearing space.
4. After long-term use, natural wear and tear is a normal phenomenon. After one year of use, if abnormal noise is found in the motor, the heating system is abnormal, the bearings in the transmission part are worn, the belt is loose or cracked, the heating constant temperature is abnormal, and the electronic control components fail, etc.
