The five step manual washing method for ELISA plates involves shaking off the liquid in the wells and flipping the enzyme-linked immunosorbent assay plate vertically to quickly remove any residual liquid from the wells, avoiding cross contamination between the wells caused by wrist shaking. Use clean thick absorbent paper (not toilet paper) to dry the liquid in the hole. When tapping, the position of the absorbent paper should be changed to avoid debris residue. Add 350 μ L of detergent to each well (freshly prepared and used, PBS buffer containing 0.05% -0.2% Tween 20), fill the well but avoid overflow. Soak and shake dry, let it stand for 1-2 minutes (intermittent shaking), then shake off the liquid and pat dry again, repeat this step 3-5 times. After drying, it is necessary to quickly add subsequent reagents to avoid protein inactivation caused by the drying of the enzyme-linked immunosorbent assay (ELISA) plate, especially in high-temperature environments. Key precautions for detergent preparation: Dilute and concentrate 20 times, use non polluting distilled water, and dissolve crystals. Clamping skills: moderate strength, avoid breaking Flat noodles or uncleanness. Avoid contamination: High concentration pore liquids may contaminate adjacent pores, so extra caution should be taken during the first two washes.
