Pharmaceutical Encyclopedia | Material Selection and Safety Analysis of Medical Eye Drops Bottles
Date: 2025-12-03Read: 5
Medical eye drops are drug preparations that are directly dripped into the mucous membrane of the eye. Their packaging (eye drop bottle) serves as the "first barrier" in direct contact with the human mucous membraneMaterial safety, compatibility, and functionalityIt is directly related to the stability of drug efficacy, patient medication safety, and user experience. Compared to oral medication packaging, eye drop bottles face more stringent challenges:
  Mucosal irritationThe thickness of the ocular mucosa is only 1/10 of the skin, and it is extremely sensitive to chemicals such as plasticizers and residual monomers. Material migration may cause redness, swelling, and stinging;
  Aseptic requirementsEye drops are mostly sterile preparations (such as single dose packaging) or non sterile preparations (requiring preservation), and the packaging needs to withstand sterilization processes (such as wet heat sterilization, irradiation sterilization) and maintain a sterile barrier;
  Dose accuracyThe single dose is only 10-50 μ L (about 2-10 drops), and the bottle mouth design should avoid the drug hanging on the wall, uneven dripping speed, or secondary pollution;
  Stability guaranteeIt is necessary to block moisture, oxygen, and light (especially for photosensitive drugs such as levofloxacin) to prevent drug oxidation, hydrolysis, or potency reduction.
Therefore, the material selection for eye drop bottles should revolve aroundBiosafety, chemical compatibility, physical functionality, sterilization compatibilityThe four core areas are developed, and strict quality control is implemented to ensure safety throughout the entire lifecycle.
1Eye drops bottleComparison of Common Materials and Characteristics
At present, the mainstream materials for medical eye drops bottles includeLow density polyethylene (LDPE), polypropylene (PP), polyester (PET), cyclic olefin copolymer (COC/COP)andglassThe comparison of its characteristics is as follows:

material
生物安全性
Chemical compatibility
Physical functionality
Sterilization compatibility
Typical application scenarios
LDPE
High inertness, no allergenicity; But the moisture permeability is high (2~5g/(m ² · d)), and long-term storage is prone to moisture absorption.
Resistant to weak acids and bases, not resistant to aromatic hydrocarbons and ketone solvents (such as ethanol containing eye drops that may swell).
Soft and easy to squeeze, easy to control drip speed; But it has low transparency (foggy) and is prone to deformation (at high temperatures).
Resistant to ethylene oxide sterilization and gamma radiation (≤ 25kGy); Not resistant to moist heat sterilization (softening at>60 ℃).
Non sterile eye drops (such as artificial tears), large capacity packaging (>10mL).
PP
BPA free, plasticizer free, strong chemical resistance (stable at pH 1-14); High melting point (160-170 ℃), stable at high temperatures.
Resistant to most solvents (including ethanol, glycerol), suitable for alcohol containing or hypertonic eye drops (such as tobramycin dexamethasone eye drops).
Higher hardness than LDPE, good drop resistance; Medium transparency (foggy), can be improved by biaxially stretching (BOPP) to enhance transparency.
Resistant to moist heat sterilization (121 ℃/30min) and irradiation sterilization (≤ 50kGy); No dissolved substances.
Aseptic/non sterile eye drops (such as antibiotics) and compound preparations that require high-temperature sterilization.
PET
Low toxicity, compliant with FDA 21 CFR 177.1630; But it has strong moisture absorption (0.2~0.5g/(m ² · d)) and needs to be stored dry.
Resistant to dilute acid and alkali, not resistant to strong acid and alkali (such as pH<3 or>9, which may hydrolyze); Sensitive to alcohols (high concentration ethanol easily swells).
High transparency (>90%), easy to observe the status of the medication; Good barrier properties (oxygen permeability ≈ 0.05cm ³/(m ² · d · atm)).
Resistant to radiation sterilization (≤ 30kGy); Wet heat sterilization is prone to deformation (glass transition temperature ≈ 70 ℃).
Photosensitive drugs (such as vitamin A palmitate eye drops) require packaging with high transparency.
COC/COP
Ultra high purity (metal ion residue ≤ 1ppm), no cytotoxicity; Strong inertia, almost no migration.
Resistant to polar/non-polar solvents (such as ethanol, dimethyl sulfoxide), suitable for complex prescriptions (such as liposomes, nanoparticle eye drops).
Transparency close to glass (>92%), low refractive index (1.53 vs 1.52 for glass); Low hygroscopicity (<0.1g/(m ² · d)).
Resistant to high temperature sterilization (180 ℃ dry heat sterilization) and irradiation sterilization (≤ 100kGy); No degradation.
Biological agents (such as antibody eye drops), innovative drugs (such as gene therapy eye drops).
Glass (borosilicate)
Good chemical inertness, no migration risk; But it is heavy, fragile, and has high transportation costs.
Resistant to all drugs, including strong acids, bases, and organic solvents; However, attention should be paid to the soluble barium and arsenic in the glass (which must comply with USP Class VI).
High transparency (>95%), resistant to high-pressure sterilization; But it is prone to detachment (silicate particles falling off).
Resistant to moist heat sterilization (121 ℃/30min) and dry heat sterilization (200 ℃); Surface silicification treatment is required to prevent detachment of the film.
High value drugs (such as biologics) and classic formulations that require long-term stability (such as atropine eye drops).
2、 Core considerations for material selection
The selection of materials for eye drops bottles needs to be comprehensively consideredDrug characteristics, packaging form, sterilization method, and patient needsSpecifically, as follows:
  1. Drug characteristics: compatibility and stability matching
  Drug pH and solventAcidic drugs (such as vitamin C eye drops, pH 3-4) should use acid resistant materials (such as PP, COC); Ethanol containing eye drops (such as some disease resistant ones) should avoid LDPE (swelling) and prioritize PP or COC;
  photosensitivityPhotosensitive drugs (such as latanoprost eye drops) should use brown PET or opaque COC (light transmittance ≤ 5%), or add a light shielding layer to the outer packaging;
  osmotic pressureHigh permeability eye drops (such as mannitol eye drops) require materials that are resistant to osmotic pressure (such as PP, COC) to avoid deformation of LDPE due to water absorption;
  biologicalsProtein and nucleic acid drugs (such as recombinant human epidermal growth factor eye drops) are prone to adsorption or inactivation on material surfaces, and low adsorption materials (such as COC and silica glass) should be selected.
  2. Packaging form: Suitable dosage and sterile requirements
  Single dose packaging (0.4~1mL)Must be sterile and preservative free, and the material must be resistant to radiation sterilization (such as PP, COC); The bottle design is "easy to break ampoule" or "pre sealed" to avoid secondary contamination;
  Multi dose packaging (5-15mL)Preservatives (such as benzalkonium chloride) need to be added, and the material should be resistant to corrosion by preservatives (such as PP being better than LDPE); The bottle mouth should be designed with a "one-way valve" or "duckbill valve" to prevent air backflow from contaminating the medicine solution;
  Large capacity packaging (>20mL)Commonly used in hospital settings (such as washing liquids), LDPE (low cost, easy to squeeze) can be selected, but it needs to enhance barrier properties (such as composite aluminum film).
  3. Sterilization method: Ensure process compatibility
  Wet heat sterilization (121 ℃/30min)Only applicable to PP and glass, it is necessary to verify the dimensional stability of the material under wet and hot conditions (such as shrinkage rate ≤ 1%) and the absence of precipitates;
  Radiation sterilization (25~50kGy of gamma rays)PP, COC, LDPE are suitable, and the molecular weight change of the material after irradiation (such as a decrease of ≤ 10% in the molecular weight of PP) and the presence of free radical residues (such as no residue in COC) need to be detected;
  Ethylene oxide (EO) sterilizationAll materials are applicable, but EO residue (≤ 4mg/device) should be controlled to avoid EO migration to the drug solution (especially for children);
  Aseptic filling (no need for sterilization)The materials need to be produced in a dust-free workshop (Class A clean area) and the surface needs to be treated with plasma to improve cleanliness.
  4. Patient Experience: Optimizing User Convenience
  Drip rate controlThe inner diameter of the bottle mouth needs to be precise (such as 0.8~1.2mm), and the "microfluidic design" ensures uniform volume per droplet (error ≤± 10%);
  Anti pollution designThe bottle mouth can be integrated with a "flip cover" or "sensing opening" (such as infrared sensing) to avoid contamination caused by finger contact;
  Grip comfortThe bottle body should conform to ergonomics (such as a diameter of 28-32mm), and the surface should be sandblasted to prevent slipping;
  Child safetyOptional "anti opening cover" (requires both hands to press and rotate) to prevent children from accidentally touching.
3、 Security analysis: full chain control from "material migration" to "biocompatibility"
The safety of eye drops bottles needs to be ensured throughMaterial migration testing, biocompatibility evaluation, sterilization validation, and clinical application monitoringFour step verification to ensure no risk.
  1. Material migration testing: controlling the leaching of harmful substances
  Simulated liquid selectionSelect simulated solution based on drug pH (e.g. 0.1mol/L HCl for acidic drugs and pH 7.4 PBS for neutral drugs);
  test conditionsAccording to the ICH Q3D guidelines, detect migrating substances (such as monomers, additives, heavy metals) under accelerated testing (40 ℃/75% RH, 6 months) or long-term testing (25 ℃/60% RH, 24 months) conditions;
  requirement:
Heavy metals (Pb ≤ 1ppm, Cd ≤ 0.1ppm);
Monomer residue (such as ethylene glycol in PET ≤ 0.5%, propylene in PP ≤ 0.1%);
Plasticizers (such as phthalates not detectable);
Total amount of dissolved substances (≤ 10mg/container).
  2. Biocompatibility evaluation: compliant with ISO 10993 standard
  cytotoxicityUsing L929 mouse fibroblasts, the cell survival rate was detected by MTT assay to be ≥ 70% (no cytotoxicity);
  Skin/mucosal irritationThe Draize test score for rabbit eyes is ≤ 1 (non irritating);
  allergenicityNo positive reaction (no sensitization) was observed in the Guinea Pig Large Scale Test (GPMT);
  GenotoxicityAmes test (bacterial recovery mutation) has no mutagenicity.
  3. Sterilization validation: Ensure the effectiveness of sterile barriers
  Sterility TestAccording to General Rule 1101 of the Chinese Pharmacopoeia, samples were cultured using membrane filtration method (sterile growth for 14 days);
  Sterilization process confirmationProve through heat distribution test (moist heat sterilization) or dose distribution test (irradiation sterilization) that sterilization parameters (such as temperature, time, dose) can kill all microorganisms (including spores);
  Integrity of sterile barrierUse dye penetration method (such as methylene blue solution) or pressure decay method to verify that the bottle body has no microcracks (leakage rate ≤ 0.01mbar · L/s).
  4. Clinical application monitoring: post marketing risk control
  Adverse reaction trackingCollect adverse reactions such as eye irritation and allergies related to eye drops bottles through the national adverse drug reaction monitoring system (such as the China ADR Monitoring Center), and analyze the causes (such as material migration and sterilization residue);
  Stability reassessmentRegular spot checks (e.g. every 2 years) are conducted on products already on the market to test the drug content, related substances, and packaging material properties (such as a decrease in barrier properties of ≤ 10%).
4、 Development Trend: From "Safety Compliance" to "Functional Innovation"
With the innovation of ophthalmic drugs (such as biologics, gene therapy) and the increasing demand for medication experience from patients, the material of eye drop bottles is moving towardsHigh barrier, low adsorption, intelligent, and greenDirection of development:
  1. High Performance Materials: Dealing with Complex Drugs
  COC/COPDue to its ultra-high purity and low adsorption, it has become the "golden material" for biologics eye drops (such as Novartis' Lucentis) ® (Leizhu monoclonal antibody) eye drops are administered in COC bottles;
  Multi-layer composite filmFor example, PP/aluminum foil/PE composite bags are used to enhance barrier properties (oxygen permeability ≤ 0.001cm ³/(m ² · d · atm)) for compound eye drops that require long-term storage.
  2. Intelligent packaging: improving medication adherence
  Electronic tags (RFID)Integrated temperature and opening time monitoring functions, prompting patients with "expired medication" or "abnormal storage temperature";
  Dose memory functionRecord the time of each medication drop through a pressure sensor, and the APP will provide synchronized reminders (such as "It has been 4 hours since the last medication, can be used again").
  3. Green materials: practicing sustainable development
  Recyclable single materialFor example, 100% PP bottles (to avoid the problem of recycling multi-layer composite materials) can improve the recycling rate through the "bottle cap integrated design";
  Biobased plasticsSuch as PLA (polylactic acid) eye drops bottles (to verify their compatibility and degradation rate with drugs), reducing dependence on petroleum based materials.
  ConclusionThe material selection of medical eye drops bottles is a balanced art of "safety, functionality, and economy", which should focus on drug characteristics, combine material science with clinical needs, and ensure safety through full process quality control. In the future, with breakthroughs in new materials and technologies, eye drop bottles will be further upgraded to be "safer, smarter, and greener", providing solid guarantees for the safety and accessibility of ophthalmic drugs.
