High resolution micro Raman spectrometer is a precision analytical instrument widely used in materials science, chemistry, biomedical and other fields to study the molecular structure and chemical composition of substances. To ensure the stable operation of the instrument and the accuracy of experimental results, a series of precautions should be followed during operation. This article integrates multiple reliable sources and summarizes the key usage points of high-resolution micro Raman spectroscopy.
1、 Preparation before the experiment
1. Instrument status check: Before starting up, it is necessary to confirm that all components of the instrument are securely connected, including the laser, sample stage, and detector, and check whether the power and signal lines are intact. At the same time, ensure that the surrounding environment of the instrument is clean and free of debris blocking heat dissipation or light paths, and avoid operating in humid, high-temperature, or strong magnetic field environments to prevent affecting detection accuracy.
2. Sample preparation: Select a suitable container based on the properties of the sample. For example, solid samples can be fixed on glass slides, while liquid samples require the use of capillary tubes or quartz colorimetric dishes, and ensure that there are no bubbles; The gas sample should be sealed in a sealed sample pool. The surface of the sample should be kept smooth to avoid interference from pollutants on the spectral quality.
3. Parameter pre setting: Based on experimental requirements, preliminarily set parameters such as laser wavelength, power, and integration time. It is recommended to initially use a lower laser power and gradually adjust it to avoid sample damage, while selecting an appropriate wavelength range and scanning frequency to balance signal strength and measurement time.
2、 Precautions during operation
1. Optical path calibration and focusing:
When starting the instrument, turn on the power in sequence, and after the self-test is completed, calibrate the optical path using a standard sample (such as a silicon wafer). By adjusting the objective lens and focusing system, ensure accurate laser beam incidence on the sample and optimize the scattered light collection path.
Calibration includes wavenumber and intensity calibration, which need to be performed regularly to maintain data accuracy. For example, adjusting the peak shift through software to ensure that the spectral characteristic peaks are consistent with the standard values.
2. Key points of data collection:
Set parameters such as spectral range, integration time, and scanning frequency in the software. A short integration time may lead to weak signals, while a long time can easily cause thermal damage to the sample; Scanning frequency can improve signal-to-noise ratio, but time cost needs to be balanced.
Maintain environmental stability during measurement and reduce vibration and noise interference. For easily photodegradable samples, priority should be given to using low-power lasers and shortening the exposure time.
3、 Post experimental processing and maintenance
1. Data saving and instrument shutdown: After completing the measurement, promptly save the spectral data and shut down the equipment in the correct order (such as shutting down the laser first, then shutting down the host and computer). Check whether the sample has been damaged due to laser irradiation, such as photolysis or thermal denaturation.
2. Daily maintenance:
Optical system: Regularly clean the lens and reflector, use dry nitrogen or specialized cleaning agents to remove dust and fingerprints; Avoid touching the grating or lens directly, and contact professional personnel if necessary.
Environmental control: The laboratory needs to maintain a constant temperature (recommended 24-26 ℃), dryness (humidity below 65%), and dust prevention, avoid direct sunlight on the instruments, and ensure good ventilation.
Computer system: Use a pure English operating system to avoid installing unrelated software; Priority should be given to using CDs for data transmission to reduce the risk of viruses.
4、 Safe and standardized operation
1. Safety protection: Wear goggles and gloves during operation to avoid direct laser beam on the eyes or skin; Do not open the casing to contact internal components while the instrument is running.
2. Training and supervision: New users need to receive technical training from the manufacturer and be familiar with the operating procedures before they can operate independently; Abnormal situations (such as errors or data anomalies) should be immediately shut down and management personnel should be contacted.
By following the above precautions, the service life and experimental reliability of high-resolution micro Raman spectrometer can be significantly improved, ensuring the acquisition of high-quality spectral data.
