1、 Basic requirements for sterilization technology
The success rate of plant tissue culture highly depends on the sterile environment, and sterilization equipment needs to meet three core indicators: sterilization efficiency (killing more than 99.9% of microorganisms), material compatibility (not damaging the composition of the culture medium), and operational safety. High pressure steam sterilization (maintained at 121 ℃ and 0.1MPa for 15-30 minutes) is still a reliable physical sterilization method, which works by denaturing microbial proteins through high temperature and high pressure. For heat sensitive instruments, ethanol immersion or ultraviolet irradiation can be used as auxiliary methods.
2、 Mainstream equipment types and technical features
1. High pressure sterilization pot
Vertical and horizontal structures are the main types, with capacities ranging from 20L to 500L. The new generation of equipment is equipped with an intelligent control system that can automatically record sterilization curves and generate reports. Key improvements include:
Pulsating vacuum technology (improving steam permeability)
Optimization of drying program (reducing residual condensed water)
Energy saving design (waste heat recovery system reduces energy consumption by 30%)
2. Filtration and sterilization device
For thermally unstable components such as hormones and antibiotics, a positive pressure filtration system using a 0.22 μ m microporous membrane is employed. Attention should be paid to the selection of filter membrane material (polyether sulfone membrane is superior to nitrocellulose membrane) and integrity testing (bubble point method verification).
3. Ozone/UV combination system
Suitable for air sterilization in vaccination rooms, UV-C band (253.7nm) destroys microbial DNA, and ozone generator (concentration ≥ 10ppm) enhances blind spot treatment capability. Need to cooperate with delayed start design to avoid personnel exposure risks.
