The Escherichia coli tube centrifuge is a new model independently developed by our company (with a speed adjustable from 0-15000 r/min), and the lower damping adopts domestic invention technology that does not require grease lubrication, solving the important problem of grease contaminated materials. All equipment is made of stainless steel, with exquisite appearance, low noise, and good separation effect. It is currently the most commonly used equipment in the biopharmaceutical industry. The operating procedures involve multiple steps and precautions to ensure efficient and safe use of the equipment.
Main characteristics of Escherichia coli tube centrifuge
High separation factor: Tube centrifuge is a separation equipment with high centrifugal force in industrial production centrifuges in China, suitable for tasks that require high centrifugal force separation.
Wide applicability: Suitable for materials with solid content less than 5% and particle diameter greater than 1-2 microns; It is also suitable for clarifying suspensions and separating emulsions with small differences in solid-liquid phase density.
Operating specifications for Escherichia coli tube centrifuge
1. Material pretreatment:
Dilution and temperature regulation: Control the temperature of the fermentation broth at 20-40 ℃ (preferably 30 ℃) to prevent low temperature from increasing viscosity or high temperature from causing antibiotic degradation.
• Filtering and impurity removal: Pre filter with a 200 mesh filter cloth to remove large particles of impurities (such as culture medium residue and mycelial clumps) and prevent blockages in the internal flow channels of the drum.
Adjust pH value: Adjust the pH of the fermentation broth to near the isoelectric point (such as pH 4.5~5.0 for some bacteria) based on the characteristics of the bacterial cells, promote bacterial aggregation, and improve separation efficiency.
2. Equipment inspection:
Sealing of the drum: Check if the sealing ring of the drum end cover is worn, ensure good sealing, and avoid liquid leakage that may cause unstable speed.
Transmission system: Check the belt tension and bearing lubricating oil level to ensure the normal operation of the transmission system.
Sensor calibration: Run idle for 5 minutes after starting up to confirm that the displayed values of the speed sensor and temperature sensor are normal.
3. Feed and acceleration:
Low speed feeding: Turn on the feeding pump and increase the centrifuge speed to 2000-3000rpm to evenly distribute the fermentation broth along the inner wall of the drum, avoiding imbalance caused by high-speed feeding.
• Gradually increase speed: After the feed is stable, gradually increase the speed to the target value at a rate of 500rpm/min.
4. Operation monitoring:
Key parameter recording: Monitor parameters such as drum speed, feed flow rate, bearing temperature, and vibration amplitude to ensure they are within normal ranges.
• Exception handling: If there is abnormal vibration or parameter abnormality, the speed should be immediately reduced and the machine should be stopped to check and handle the problem.
