Process principle of ultrasonic cell disruptor
Date: 2024-09-14Read: 5
Ultrasonic cell disruptor is a device that can perform cell lysis through ultrasound technology, mainly used for cell analysis, virus research, and other fields in biological sciences, medical research, and laboratories.
Ultrasonic cell lysis is the use of high-energy ultrasound oscillation to destroy molecules and structures within cells. High intensity ultrasound forms cavitation bubbles in liquid. This kind of foam grows and collapses during ultrasonic vibration, forming a tiny fluid flow dynamics, and processing biological substances into very small fragments. These fragments can be used for further research, such as isolating proteins, DNA, or RNA, and conducting further analysis.
Ultrasonic cell disruptors typically use an elongated clay compression head to place the crushed sample and use water or shock-absorbing materials to improve the stability of the oscillating mechanical system. Before the experiment, standardization is required to ensure high accuracy when calibrating the equipment based on the biological characteristics and quantity of the sample.

The process of cell lysis generally includes the following steps:
1. Place cells or tissues in a fragmented sample container;
2. Add solvents (such as deionized water, buffer solution, and detergent) to disturb the cell membrane and cell wall;
3. Prepare the crusher and injection head before crushing, ensuring a stable connection between the injection head and crushing head;
4. Adjust various parameters of the machine, including crushing time (the longer the time, the more times it will be crushed), power, oscillation intensity, and solvent concentration;
5. Perform pre fragmentation steps to ensure that dead cells have been processed and to avoid their substances affecting subsequent fragmentation;
6. Start to break down and destroy other structures of the cell, producing fragments;
7. Stop ultrasound, separate the top solvent and the overall crusher and fragments;
8. Collect the required crushed components.
