Analysis of the specific workflow of atomic absorption spectrometer
Date: 2025-12-09Read: 6
Atomic absorption spectrometer is an instrument that utilizes the high absorption of specific wavelengths of light by atoms for quantitative analysis of elements. The core idea is to convert the metal elements in the sample into atomic vapor in a single atomic state (usually generated in flames, graphite furnaces, or inductively coupled plasmas), and then let monochromatic light or narrowband light sources pass through the atomic vapor. The atoms of the element to be tested have strong absorption of radiation at characteristic wavelengths in their ground or low excited states, and the amount of light absorbed is proportional to the concentration of the element in the sample, thus achieving quantitative analysis.

  Atomic absorption spectrometerThe workflow:
1. Method development and light source selection
-Select the appropriate light source (such as sodium lamp, magnesium lamp, etc.) and wavelength based on the target element. Different elements have maximum absorption coefficients at specific wavelengths, which need to be determined in advance.
2. Sample preparation and pretreatment
-Using steps such as digestion, complexation, and dilution, the solid sample is converted into a transparent liquid to ensure that it is within the linear range of analysis.
-Be careful to avoid pollution, element migration, and chemical interference. Common strategies such as internal standards and matrix matching are used to improve precision.
3. Atomization and Measurement
-Introduce the sample into a flame or graphite furnace to form atomic vapor.
-The specific wavelength light emitted by the light source passes through the atomic vapor, and the unabsorbed light enters the detector through transmission; The stronger the absorption, the weaker the detection signal, reflecting the concentration of elements in the sample.
4. Data analysis and result output
-Establish a standard curve using the standard addition method or external standard method to correct background signals and interference.
-Based on the calibration curve, calculate the concentration of elements in the sample, provide measurement values and uncertainties, and conduct quality control evaluations.
Application areas of atomic absorption spectrometer:
1. Environmental analysis
-Detection of trace metal elements such as lead, mercury, arsenic, cadmium, chromium, nickel, copper, zinc, etc. in water bodies, soils, and sediments. AAS is widely used in water quality monitoring, wastewater discharge, and soil pollution assessment.
2. Food and Agriculture
-Trace metal content, elemental nutrition assessment, and pollutant monitoring in food. Correction of matrix complexity is required to ensure minimal interference of food matrix on analysis.
3. Biological samples and clinical analysis
-Quantification of metal elements in biological fluids (blood, urine, tissue) for diagnosis, drug metabolism research, and exposure assessment. Strict pre-processing and internal standard strategies are usually required.
4. Industrial and Material Analysis
-Trace element analysis in metal materials, alloys, and coatings to evaluate material quality, process control, and corrosion resistance.
