Common problems and solutions of diagnostic reagent filling machine
Date: 2025-05-27Read: 5
The following is a systematic analysis and solution to common problems with diagnostic reagent filling machines, summarized and organized based on industry practice and equipment characteristics:
Common problems and solutions of diagnostic reagent filling machine
1、 Abnormal filling accuracy
Phenomenon:
The fluctuation of filling volume exceeds the allowable deviation (such as ± 1% → ± 5%)
The infusion volume between reagent bottles is significantly uneven
Solution:
1. Servo system calibration: Re calibrate the step angle of the servo motor to ensure the accuracy of the piston motion stroke.
2. Liquid characteristic compensation: Adjust the filling head delay closing time based on the viscosity of the reagent (especially high viscosity magnetic particle reagents).
3. Pipeline optimization: Check and replace aging silicone tubes (recommended to replace every 2 million filling cycles) to eliminate wall adsorption effects.
2、 Liquid residue/cross contamination
Phenomenon:
Droplets formed by liquid hanging on the outer wall of the filling needle
Cross contamination detected by different batches of reagents
Solution:
1. Anti hanging liquid design: using hydrophobic coating filling needle (contact angle>110 °), combined with positive pressure air knife blowing device.
2. Cleaning program upgrade:
Perform a three-level cleaning process: rinse with pure water → soak in 1% NaOH → dry with sterile compressed air.
Increase the frequency of CIP cleaning (mandatory cleaning after each batch).
3. Isolation measures: Install laminar flow hoods in key areas and use independent isolators for filling high-risk reagents.
3、 Equipment blockage/stuck bottle
Phenomenon:
The filling needle is blocked by reagent crystallization
The positioning deviation of the penicillin bottle caused the mechanical arm to collide
Solution:
1. Intervention of temperature control system: Install a temperature control module (adjustable from 4-25 ℃) in the filling area to prevent crystallization of low-temperature reagents.
2. Visual positioning correction: Install a CCD camera to real-time detect the position of the bottle mouth and dynamically adjust the coordinates of the robotic arm.
3. Dumb proof design: Install pressure sensors on the bottle feeding track to detect abnormal resistance and immediately trigger an emergency stop.
4、 Sealing issue
Phenomenon:
Micro pores appear on the aluminum foil seal (affecting the stability of the reagent)
Uneven sealing force of rubber plug
Solution:
1. Sealing parameter optimization: Adjust the heat sealing temperature (120-180 ℃) and pressure (0.3-0.6MPa) according to the thickness of the aluminum foil (usually 0.2-0.5mm).
2. Sealing detection: Integrated laser headspace analyzer, real-time monitoring of oxygen permeability after sealing (should be<0.01cc/pkg · day).
3. Consumables management: Establish a database of rubber plug deformation and force the replacement of O-rings every 5000 presses.
5、 Data traceability defects
Phenomenon:
Production batch and filling data matching error
Key parameters not fully recorded
Solution:
1. RFID binding system: Implant chips at the bottle entry station to achieve single bottle level data traceability.
2. Audit tracking enhancement: Enable electronic signature function to record metadata such as operator ID and parameter modification timestamp.
3. Database redundancy backup: Adopting a dual server hot backup mode to ensure compliance with 21 CFR Part 11.
