If the results do not meet the set standards, the Xcalibur sequence will automatically pause for users to investigate possible causes and avoid wasting the remaining samples.

The adaptive retention time function can automatically adjust the retention time of the analyte based on the collected reference spectrum file, reducing missing values and narrowing the retention time window for target quantitative analysis, effectively improving analysis throughput. Can be used for PRM quantification or SQUAD and hybridDIA methods.
In addition, the addition of AdaptiveRT can better achieve the complete process from discovery to target analysis in a single system.

The HybridDIA method combines non target DIA analysis and target based tMS2 analysis in the same injection, allowing for simultaneous discovery type DIA and target quantification. It is suitable for focusing on specific target objects while non target analysis.
In the Ascend instrument, parallel analysis can be better achieved by using both Orbitrap and ion trap mass analyzer. Orbitrap is used for DIA scanning and ion trap is used for targeted MS2 scanning, achieving high sensitivity and high-throughput quantification of specific targets without sacrificing DIA depth coverage performance.

The RTS SPS MS3 method can significantly improve quantitative accuracy in TMT experiments, and the newly added signal-to-noise ratio threshold filtering can further enhance quantitative credibility.
When real-time retrieval (RTS) is enabled and the Closeout function is enabled in TMT SPS MS3 mode, the interface will display the newly added 'MS3 signal-to-noise ratio threshold' option, which can be customized. If the MS2 spectrum identified through real-time search generates a low-quality MS3 spectrum, it will no longer be included in the quantitative values of the corresponding proteins, which is more helpful in improving the accuracy of quantification.
Update: Real time spectral library retrieval RTLS

Real time library search (RTLS) can achieve intelligent collection of similar compounds in the database. In the small molecule analysis mode, the newly added Modified Cosine Similarity takes into account the mass deviation between parent ions when comparing experimental spectra and library spectra, and corrects the mass to charge ratio of fragment ions accordingly, thereby calculating similarity more accurately.
Using the updated RTLS, no manual intervention is required, and structurally related compounds can be automatically discovered through library searches. And due to higher matching scores, more matching compounds can be discovered in unknown samples.
Update: Faster ETD response time

ETD fragmentation has important applications in various research fields, including glycosylation modification, disulfide bonding, de novo sequencing, top-down, middle down analysis, and so on. Among them, ETD reaction time is a very important parameter that affects fragmentation efficiency, overall cycle time, and so on.
After the update, the ETD reaction time in the ion trap is shortened, closer to the ion implantation time, which can achieve better parallelization efficiency.
