Chemical modification effects: Covalent binding between epoxy groups and enzymes may disrupt their active centers (such as heme cofactors), leading to a decrease in catalytic efficiency. Environmental sensitivity: Although PEG chains can protect enzymes from protease degradation, high temperatures or organic solvents may damage the tertiary structure of enzymes, requiring strict control of storage conditions (such as low temperatures and neutral pH).
PEG chain detachment or degradation
Hydrolysis or enzymatic hydrolysis: PEG chains may break due to in vivo esterase or hydrolysis, leading to ineffective modification. For example, in long-term circulation, PEG may be metabolized by the liver or kidneys, shortening its half-life.
Crosslinking side reactions: Epoxy groups may undergo non-specific crosslinking with other molecules (such as plasma proteins), forming aggregates that affect drug distribution and clearance.
