  Pre filled syringe filling machineThe material properties directly affect the safety of drugs and the stability of equipment. It is necessary to consider corrosion resistance, cleanliness, and material adaptability, especially in the fields of biologics, vaccines, etc. The material selection must comply with the ISO 10993 biocompatibility standard. ​
The core contact components are preferably made of 316L stainless steel, which contains 2-3% molybdenum and has better intergranular corrosion resistance than 304 stainless steel. It can withstand acidic and alkaline environments with pH 1-14 and is suitable for filling corrosive drugs such as amino acid infusion solutions and heparin sodium. The surface needs to be treated with electrolytic polishing, with a roughness Ra ≤ 0.8 μ m, to reduce drug residue and microbial growth, while avoiding metal ion precipitation (migration amount ≤ 0.1mg/kg). The filling needle is made of ceramic material (zirconia) with a hardness of HRC 85, which not only solves the wear problem of stainless steel needles (prolongs their service life by three times), but also avoids chemical reactions with sulfur-containing drugs (such as cephalosporins). ​
The material of the sealing system needs to be adapted specifically. Nitrile rubber (NBR) sealing rings are suitable for non-polar solvents (such as vegetable oil injection), but they are prone to swelling in polar drugs (such as water-based vaccines) and need to be replaced with fluororubber (FKM). Its temperature resistance range is -20 ℃ to 200 ℃, and it has excellent compatibility with alcohols and esters. The O-ring of the piston drive mechanism is made of silicone rubber (VMQ), which has passed USP Class VI certification and can directly contact blood products. The compression long-term deformation rate is ≤ 10%, ensuring stable long-term sealing performance. ​
The material selection of non-contact components needs to balance functionality and cost. The equipment frame is made of 304 stainless steel, which has been acid washed and passivated to meet the anti rust requirements of GMP for clean area equipment; The surface of the conveyor track is coated with PTFE coating, with a friction coefficient as low as 0.04, reducing the risk of scratches during the pushing of pre filled syringes. It also has chemical resistance and can withstand long-term wiping with 75% alcohol for disinfection. The electrical component casing is made of ABS engineering plastic, with a surface finish of ▽ 6, which is easy to clean and has reliable insulation performance (breakdown voltage ≥ 20kV/mm). ​
Material compatibility needs to be verified through rigorous testing. For protein drugs (such as monoclonal antibodies), it is necessary to test the adsorption rate of the stainless steel surface on the drug (requirement ≤ 0.1%); When filling drugs containing surfactants (such as liposomal vaccines), it is necessary to verify the dissolution of rubber seals (total organic carbon ≤ 0.5mg/L). Adopting simulated filling test: using a 50% ethanol solution to circulate and contact the equipment for 24 hours, detecting the heavy metal content (total amount of lead and cadmium ≤ 0.1ppm) in the leachate, and observing whether the material has deteriorated such as cracking and discoloration. ​

Customized material schemes are required for the filling of special materials. When filling traditional Chinese medicine extracts, due to the presence of tannins, the inner wall of the filling chamber needs to be changed to Hastelloy C276, which is resistant to various organic compounds and does not react with polyphenolic substances; For low-temperature filling (such as biological agents at -5 ℃), replace the plastic bearings of the transmission components with polyetheretherketone (PEEK) to avoid low-temperature embrittlement (embrittlement temperature ≤ -60 ℃). In daily maintenance, a cleaning plan should be developed based on the characteristics of the medication: acidic medication residues should be cleaned with a 0.5% sodium carbonate solution, and alkaline residues should be neutralized with a 1% nitric acid solution to prevent chronic corrosion of the material. ​
