The administration route of drugs with local and systemic effects on the skin has received considerable attention in recent years. However, the main obstacle for drugs to penetrate the skin and exert their pharmacological activity is the inherent barrier of the skin, the stratum corneum (SC). The stratum corneum is the topmost layer of the skin, composed of keratinocytes filled with keratin and intercellular lipids. When conducting drug and cosmetic transdermal diffusion tests (IVPT) and in vitro efficacy evaluation of 3D epidermal models, visual and electrical impedance values (Trans Epidermal) are used Electrical Resistance (TEER) and Transdermal Water Loss (TEWL) were used as indicators to evaluate the functional integrity of in vitro skin slice models such as pig skin, mouse skin, and cultured skin. Skin model impedance testerThe operational process
Slice the experimental skin to be tested (such as pig skin, mouse skin, and other animal ex vivo skin used in experiments, 3D recombinant epidermal models, etc.) between two chambers (such as the supply and receptor cells of the Franz diffusion cell), inject 0.9% NaCl solution into each chamber, and insert silver chloride electrodes into each chamber to measure the impedance between the electrodes. The evaluation method varies depending on the cells used. As a preparation for transdermal absorption experiments, the characteristics of the skin membrane used are evaluated and screened based on the results of electrical impedance measurements. In addition, stratum corneum cells with reduced barrier function often have lower impedance, and the biological skin barrier function is evaluated based on the electrical impedance of water and stratum corneum.
