The stratum corneum (SC) is composed of highly organized and structured layers of dead cells, which are crucial for maintaining the water gradient between the upper layers of the skin. Many factors can interfere with the moisture barrier of the skin, which may cause dryness, irritation, or itching, thereby affecting the overall quality of the skin. Tape peeling technology has been established as a simple technique for sampling or evaluating the function of the epidermal stratum corneum (SC), and has been widely used in dermatology research. SquameScan 850A generates a circular beam (wavelength=850 nm) that passes through a tape strip containing SC and measures the decrease in light intensity (as a percentage relative to the initial intensity) in an area with a diameter of 13mm. The large area of measurement is a significant advantage, as the uneven distribution of keratinocytes on the tape is balanced. Compared with the measurement at 430nm, the error caused by molecular absorption is basically negligible at the measurement wavelength of 850nm, and thermal denaturation is prevented. When the tape strip is placed on the measuring head, the absorption value will be immediately displayed on the instrument without damaging the sample, allowing for quick and non-destructive reading of the results.
